patterns of vascular endothelial growth factor expression in hematopoietic malignant cells

چکیده
background and objective: vascular endothelial growth factor (vegf) is a cytokine which is overexpressed in many malignant cancers including leukemia. vegf plays an important role in tumor invasion and metastasis. determination of the pattern of vegf expression in human leukemic cell lines could be useful not only in screening of new antileukemic agents but also to study the mechanism of their action. in the present study, the pattern of vegf production in some human leukemic cell lines has been assessed in vitro. materials & methods: three human leukemic cell lines (molt-4, jurkat and u937) were used in this study. the cells were cultured in complete rpmi medium and then incubated with different concentrations of phorbolmyristate acetate (pma) or phytoheamagglutinin (pha) for 48 hours. the level of vegf secreted in the cellculture supernatants were measured with enzyme-linked immunosorbent assay kits (r and d systems). results: u937 cells produced a large amount of vegf without any stimulus and pha/pma did not show any substantial effect on vegf production by u937 cells. however, molt-4 and jurkat cells, produced vegf less than u937 when cultured alone (with no stimulation) and pma/pha significantly increased the vegf production in these cells dose-dependently. conclusion: different patterns of vegf expression were shown in different leukemic cell lines (molt-4, jurkat and u937) used in this study. these cells could be useful tools for screening of vegf modulators and so for planning of new drugs for treatment of leukemia and other vegf related disorders.

متن کاملMesenchymal Stem Cells Differentiate to Endothelial Cells Using Recombinant Vascular Endothelial Growth Factor –A
Background: Vascular endothelial growth factor-A (VEGF-A), an endothelial cell-specific mitogen produced by various cell types, plays important roles in cell differentiation and proliferation. In this study we investigated the effect of recombinant VEGF-A on differentiation of mesenchymal stem cells (MSCs) to endothelial cells (ECs). Methods: VEGF-A was expressed in E. coli BL21 (DE3) and BL21...
متن کاملEvaluation of Periodontal Disease Effect on the Expression of Vascular Endothelial Growth Factor and Nerve Growth Factor in Dental Pulp
Introduction: Researchers have always been interested in the close and mutual relationship between the pulp and periodontal tissues. Although the effects of pulpal disease on periodontium have been well known, the clear relationship between periodontitis and pulp has not been documented . It seems that bacteria and inflammatory products of periodontitis can affect pulp through accessory canals,...
متن کاملExpression of Vascular Endothelial Growth Factor in Nasal Polyp and Chronic Rhinosinusitis
Background: Nasal inflammatory disorders such as chronic rhinosinusitis and nasal polyp are among the most prevalent complications with high socioeconomic costs. Vascular Endothelial Growth Factor (VEGF) plays a key role in angiogenesis and cell proliferation. In the present study the effect of VEGF on the development and prognosis of chronic rhinosinusitis and nasal p...
متن کاملP-171: Expression of Vascular Endothelial Growth Factor Receptors In Endometriosis
Background: Endometriosis is a disease which is defined by the growth of endometrium-like tissue outside of the uterine cavity. Literatures show that VEGF by interaction with their receptors, Flt-1 (Fms-like tyrosine kinase-1 or VEGFR-1) and Flk-1/KDR (fetal liver kinase/ kinase-insert domain receptor or VEGFR-2) is related to pathogenesis of endometriosis. The purpose of this study was to eval...
متن کاملP-189: Investigation of Vascular Endothelial Growth Factor Receptors Expression in Ectopic Pregnancy
Background: Ectopic pregnancy (EP) is a complication of conception in which the embryo implants outside of uterine cavity. The increasing incidence of serious maternal morbidity resulting from EP has prompted the search for biomarkers to aid in early diagnosis and take advantage of conservative treatments. One of the effective biomarkers in EP is vascular endothelial growth factor (VEGF). VEGF ...
